Senior Discovery Scientist, Synthetic Biology

Full-Time
South San Francisco

About Pattern Biosciences

Do you want to be at the forefront of a revolution in healthcare? At Pattern Biosciences, we build DNA computers that run molecular programs in cells to treat cancer and other diseases. Using multi-input molecular-level computation in individual cells, our approach leads to curative therapies where the conventional one drug-one target paradigm has failed to deliver effective treatments. We have established its headquarters in South San Francisco, and it is building a world-class team to translate two decades of progress in DNA computing and synthetic biology into the clinic.

About the Role

Are you passionate about working with cutting-edge technologies to develop next generation therapies? You will advance the early pipeline of innovative cancer therapies harnessing our biomolecular computing platform. You will engineer  genetic scaffolds to compute across multiple molecular inputs inside cells to program cancer cells for destruction with exquisite specificity. You will focus on individual sensor components, molecular computing scaffolds, and fully assembled therapeutic leads, as standalone DNA constructs and as genetic payloads of viral and non-viral delivery vectors.

What you'll do

  • Design and construct barcoded genetic component libraries, logic scaffolds, effectors, and product leads;
  • Plan and execute in vitro and in vivo experiments to generate informative and clinically-translatable datasets. The data from these experiments will be used to drive the design-build-test cycle of components and fully-assembled therapeutic leads.
  • Characterize component libraries and individual therapeutic candidates as naked and vectorized DNA in mammalian cell culture and in vivo specimens using next-generation sequencing, fluorescent microscopy, flow cytometry, qPCR, and plate reader assays;
  • Map the relationship between naked and vectorized DNA performance and assess the impact of DNA sequence on viral vector manufacturing.

What you'll bring

  • Excellent knowledge of gene regulatory and gene expression processes and mechanisms in human cells, and the ability to translate this knowledge into actionable genetic designs;
  • Deep knowledge in experimental planning and execution of high-throughput cloning of barcoded libraries and individual genetic constructs using “Golden Gate”, Gibson assembly, yeast homologous recombination, restriction-ligation;
  • Experience in mammalian cell culture, fluorescent microscopy, flow cytometry, cell sorting, plate reader assays including ELISA, and RT-qPCR assays;
  • Hands-on experience in library preparation for next-generation sequencing;
  • Manufacturing of Adeno-Associated virus (AAV) vectors;
  • Preferably, prior experience in animal experimentation.

Who You Are

  • PhD degree in biosciences including genetics, cancer biology, bioengineering and related fields;
  • 1-2 years of industry and/or postdoctoral research experience is preferred but not required;
  • Track record demonstrating creativity and passion for science and patient care;
  • Desire and ability to operate in a high growth environment and deliver results within specified timelines and budgets.

This is an in person role based at our headquarters in South San Francisco
